Isolation, characterization, and genetic complementation of a cellular mutant resistant to retroviral infection.

چکیده
By using a genetic screen, we have isolated a mammalian cell line that is resistant to infection by retroviruses that are derived from the murine leukemia virus, human immunodeficiency virus type 1, and feline immunodeficiency virus. We demonstrate that the cell line is genetically recessive for the resistance, and hence it is lacking a factor enabling infection by retroviruses. The block to infection is early in the life cycle, at the poorly understood uncoating stage. We implicate the proteasome at uncoating by completely rescuing the resistant phenotype with the proteasomal inhibitor MG-132. We further report on the complementation cloning of a gene (MRI, modulator of retrovirus infection) that can also act to reverse the inhibition of infection in the mutant cell line. These data implicate a role for the proteasome during uncoating, and they suggest that MRI is a regulator of this activity. Finally, we reconcile our findings and other published data to suggest a model for the involvement of the proteasome in the early phase of the retroviral life cycle.

Isolation and Characterization of a New Peroxisome Deficient CHO Mutant Cell Belonging to Complementation Group 12
We searched for novel Chinese hamster ovary (CHO) cell mutants defective in peroxisome biogenesis by an improved method using peroxisome targeting sequence (PTS) of Pex3p (amino acid residues 1–40)-fused enhanced green fluorescent protein (EGFP). From mutagenized TKaEG3(1–40) cells, the wild-type CHO-K1 stably expressing rat Pex2p and of rat Pex3p(1–40)-EGFP, numerous cell colonies resistant to...
متن کاملIsolation and Characterization of Microsatellite Markers from Endangered Species (Camelus bactrianus)
Iranian bactrian camel (Camelus bactrianus) is an endangered livestock breed with distribution in northwest of Iran. Microsatellites are a powerful marker for animal genetic and cell line identification and population genetic study. In this study, after producing more than 40 Camelus bactrianus fibroblast cell lines, microsatellites loci from the genome of Iranian Camelus bactrianus cell lines ...

Objective(s) Staphylococcus aureus is a common cause of human infection, and emergence of vancomycin-resistance S. aureus is a great concern for treatment of methicillin-resistant S. aureus,(MRSA) in recent years (MRSA). Here, we report the isolation of high-level VRSA. Materials and Methods S. aureus was isolated from foot ulcer of a diabetic woman in Tehran, Iran. Antibiotic susceptibilit...
متن کاملProduction of transgenic Paulownia tomentosa (Thunb.) steud. using chitosan nanoparticles to express antimicrobial genes resistant to bacterial infection
Paulownia tomentosa (Thunb.) Steud. is a very important hard woody plant, an extremely fast-growing tree and produce timber. Therefore, there is a demand to produce transgenic Paulownia plant resistant to bacterial infection. Microbial infection (especially bacterial one) is serious sever and cause a loss in plant productivity as they bear upon the character and amount of plan...
